Estimates of the rates of heterotrophic nitrogen fixation (HNF) and denitrification by acetylene reduction and acetylene inhibition, respectively, were determined for the roots and culms of wild rice, Oryza glumaepatula, in Lake Batata, a lake severely impacted by the dumping of bauxite tailings into the lake basin. Two stations were established in different parts of the lake: one in a natural area and other in an impacted area of the lake. In this regard, the aim of this study was to evaluate the agronomic efficiency (AE) of three bacterial strains isolated from acid soils with high Al content from the Amazon region in an Argissolo Vermelho Distrófico típico soil (Typic Rhodustults - USDA Classification) from the municipality of Formiga, MG, Brazil. Alfalfa yield and quality as function of nitrogen fertilization and symbiosis with Sinorhizobium meliloti Scientia Agricola

The efficiency of the symbiotic process between alfalfa (Medicago sativa L.) and Sinorhizobium meliloti can be drastically affected by soil nitrogen. This research evaluates how mineral fertilization affects the contribution of symbiosis to the yield and quality of alfalfa in two experiments under controlled conditions, and a third one in the field. Under controlled conditions, positive effect of nitrogen fertilizer was observed, whereas, in the field, negative effect of the mineral nitrogen (450 kg ha-1 year-1) on the symbiotic process was observed, with reduction of nodulation and nitrogenase activity. ABSTRACT Certain nodulating nitrogen-fixing bacteria in legumes and other nodule endophytes perform different plant-growth promoting processes. The objective of this study was to evaluate 26 bacterial strains isolated from cowpea nodules grown in floodplain soils in the Brazilian savannas, regarding performance of plant-growth promoting processes and ability to enhance cowpea growth. The biodiversity of rhizobium in soils of the São Francisco Valley is unknown and can be studied using cowpea as trap plants. The objective of this study was to verify the diversity of diazotrophic bacteria that nodulate cowpea in soils of the lower half of the São Francisco River Valley by morphological and genotypic characterization. ABSTRACT Brazil nut is a very important nontimber forest product in the Amazon region. Propagation of this tree still represents a challenge due to slow and uneven seed germination. In this context, plant growth-promoting bacteria can facilitate the process of propagation. The aims of this study were to isolate and characterize endophytic bacteria from the roots of Brazil nut trees in native terra firme forest and cultivation areas in northern Brazil, and to identify mechanisms by which bacteria act in plant growth promotion. Abstract: The objective of this work was to compare three methods of 15N isotope application to the soil in order to determine the recovery efficiency of nitrogen derived from green manure by corn (Zea mays). The used experimental design was a randomized complete block with six replicates. The treatments consisted of three 15N isotope methods: indirect method I, isotope dilution with 15N-labelled synthetic fertilizer applied through a small hole next to each corn plant; indirect method II, isotope dilution with 15N-labelled synthetic fertilizer spread in the furrow next to the corn planting row; and direct method, application of a legume straw as green manure, treated with 2.23% excess 15N. Diversity and nitrogen fixation efficiency of rhizobia isolated from nodules of Centrolobium paraense PAB

The objective of this work was to isolate and characterize rhizobia from nodules of Centrolobium paraense and to evaluate their symbiotic efficiency. Soil samples collected from four sites of the Roraima Cerrado, Brazil, were used to cultivate C. paraense in order to obtain nodules. Isolates (178) were obtained from 334 nodules after cultivation on medium 79. Twenty-five isolates belonging to six morphological groups were authenticated using Vigna unguiculata and they were characterized by 16S rRNA. Draft genome sequence of Bradyrhizobium manausense strain BR 3351T, an effective symbiont isolated from Amazon rainforest BJM

ABSTRACT The strain BR 3351T (Bradyrhizobium manausense) was obtained from nodules of cowpea (Vigna unguiculata L. Walp) growing in soil collected from Amazon rainforest. Furthermore, it was observed that the strain has high capacity to fix nitrogen symbiotically in symbioses with cowpea. We report here the draft genome sequence of strain BR 3351T. The information presented will be important for comparative analysis of nodulation and nitrogen fixation for diazotrophic bacteria. A draft genome with 9,145,311 bp and 62.9% of GC content was assembled in 127 scaffolds using 100 bp pair-end Illumina MiSeq system. The RAST annotation identified 8603 coding sequences, 51 RNAs genes, classified in 504 subsystems.

Draft genome sequence of Bradyrhizobium sp. strain BR 3262, an effective microsymbiont recommended for cowpea inoculation in Brazil BJM

Firstly, this strain was assigned as Bradyrhizobium elkanii, however, recently a more detailed genetic and molecular characterization has indicated it could be a Bradyrhizobium pachyrhizi species. Draft genome sequence of Bradyrhizobium sp. strain BR 3267, an elite strain recommended for cowpea inoculation in Brazil BJM

Abstract The strain BR 3267 is a nitrogen-fixing symbiotic bacteria isolated from soil of semi-arid area of Brazilian Northeast using cowpea as the trap plant. This strain is used as commercial inoculant for cowpea and presents high efficient in nitrogen fixation as consequence of its adaptation potential to semi-arid conditions. We report here the draft genome sequence of Bradyrhizobium sp. strain BR 3267, an elite bacterium used as inoculant for cowpea. Whole genome sequencing of BR 3267 using Illumina MiSeq sequencing technology has 55 scaffolds with a total genome size of 7,904,309 bp and C+G 63%. Annotation was added by the RAST prokaryotic genome annotation service and has shown 7314 coding sequences and 52 RNA genes.

Effect of point mutations on Herbaspirillum seropedicae NifA activity BJMBR

NifA is the transcriptional activator of the nif genes in Proteobacteria. It is usually regulated by nitrogen and oxygen, allowing biological nitrogen fixation to occur under appropriate conditions. NifA proteins have a typical three-domain structure, including a regulatory N-terminal GAF domain, which is involved in control by fixed nitrogen and not strictly required for activity, a catalytic AAA+ central domain, which catalyzes open complex formation, and a C-terminal domain involved in DNA-binding.
